TTyrosinase structural gene . Prototrophic. Multiple alleles from different original strains were distinguished electrophoretically and by thermolability. The primary structure (407 amino acids) was determined by amino acid sequencing the product of allele T^L. The polypeptide contains a cysteine and a histidine linked by a thioether bridge. Precursor contains a carboxy-terminal extension. Two forms of enzyme were demonstrated in a heterokaryon. Scored by color reaction with DL-DOPA as substrate. For regulation, see refs. Tyrosinase synthesis is induced by a variety of compounds, including cycloheximide and amino acid analogs. Sulfhydryl compounds can also affect tyrosinase expression. The addition of copper before cycloheximide induction increases the yield of enzyme. Extensive biochemical and biophysical analyses of the enzyme; enzyme crystallized. Used as a reporter gene.
